Objective
I believe that if we want to unlock the true potential of the single-cell revolution for patients, we need to find new ways to interrogate cellular states, actions and interactions at scale and in vivo. With TACO, I want to achieve this to enable efficient targeting of myeloid cells, the main effectors of central nervous system (CNS) damage in multiple sclerosis (MS). To prepare for this we have acquired unique, highly-resolved datasets that allow us to capture the signals that myeloid cells receive across disease and lesion trajectories in MS. In the following work packages (WPs), I now want to extract the cues best suited to target myeloid cells in neuroinflammation:
In WP 1, Probing, I will develop in vivo CRISPR screens that allow us to uncover in a comprehensive and unbiased manner the essential signals that instruct myeloid cell states, actions and interactions in MS models.
In WP 2, Perturbing, I will combine CRISPR manipulations with single-cell analysis, multi-photon microscopy and spatial transcriptomics to delineate how these signals define the molecular signatures of myeloid cells, their functional properties and their interaction maps in the inflamed CNS.
In WP 3, Prioritizing, I will identify the signalling pathways best suited for therapeutic intervention based on their activity across MS models, lesion stages and sites.
TACO is groundbreaking because it unlocks the potential of existing and emerging (multi)omics datasets for the design of therapeutic interventions targeted to disease stages, lesion sites and cellular states. TACO is impactful as it will provide new targets for modulating progressive CNS inflammation in MS and build a versatile pipeline that can be readily adapted to the growing number of neurological conditions to which myeloid cells contribute.
