Building a full causal Portrait of cell differentiation

Objective

Cell atlases describe what cells are, but not how they become. PowerPortrait tackles this gap by constructing an experimentally grounded and well-powered causal portrait of an entire differentiation process.
Liver macrophages offer a uniquely tractable in vivo model: I have established proof-of-concept pipelines that cover multiplexed in vivo CRISPR screens, guide-RNA capture from fixated samples, and statistical modeling of perturbed differentiation dynamics. These proof-of-concepts reveal the true complexity of differentiation - an interplay of dozens of pathways - yet demonstrate that this complexity can now be approached systematically.
Building on these insights, PowerPortrait will develop a framework for causal single-cell biology that quantifies how much of a differentiation process is currently explained, what remains unresolved, and what additional data are needed. It integrates three key advances:
(i) estimating, predicting and maximizing discovery power through generative models and sample banking (POWERFORECAST);
(ii) timing and detecting perturbations during differentiation (CONDITIONALKO); and
(iii) continuously refining the portrait by creating or profiling only the most informative samples (PORTRAITOPTIMIZER).
Together, these advances provide a generalizable blueprint for causal in vivo portraits of cell differentiation, yielding predictive, mechanistic, and regenerative insights into macrophage differentiation - one of the most versatile, disease-relevant and enigmatic immune cells.

Fields of science (EuroSciVoc)

CORDIS classifies projects with EuroSciVoc, a multilingual taxonomy of fields of science, through a semi-automatic process based on NLP techniques. See: The European Science Vocabulary.

Programme(s)

Multi-annual funding programmes that define the EU’s priorities for research and innovation.

Topic(s)

Calls for proposals are divided into topics. A topic defines a specific subject or area for which applicants can submit proposals. The description of a topic comprises its specific scope and the expected impact of the funded project.

Funding Scheme

Funding scheme (or “Type of Action”) inside a programme with common features. It specifies: the scope of what is funded; the reimbursement rate; specific evaluation criteria to qualify for funding; and the use of simplified forms of costs like lump sums.

Call for proposal

Procedure for inviting applicants to submit project proposals, with the aim of receiving EU funding.

Net EU contribution

Net EU financial contribution. The sum of money that the participant receives, deducted by the EU contribution to its linked third party. It considers the distribution of the EU financial contribution between direct beneficiaries of the project and other types of participants, like third-party participants.
