Objective
Galectins are a family of glycan binding proteins that play central roles in immunity, inflammation and cancer. They are widely recognized targets in basic research as well as in diagnostic and therapeutic development, making tools that selectively label galectins highly valuable. However, current commercial options either rely on recombinant galectins which are labelled ex situ and subsequently added into target cell lines or on antibody based assays that are excellent for in vitro quantification of galectins but do not allow direct interrogation of function and tracking of dynamics.
Within the ERC StG GlycoFold, we have designed synthetic glycans capable of selectively binding and modifying specific amino acid residues in peptides. Building on this groundwork, G EFF will develop synthetic carbohydrate reagents that i) selectively recognize galectins via their carbohydrate recognition domain and ii) catalyze their covalent modification with a small fluorescent tag at a defined site.
In contrast to available methods, G EFF will enable direct labeling of native galectins and quantitative monitoring of their biological functions in simple in vitro assays. G EFF will benefit disease oriented research, diagnostic development and basic investigations in glycobiology, while opening a new technological space for glycan guided protein editing.
