Iterative Pi-Guided Polyaromatic Synthesis

Objective

Polyaromatics are ubiquitous structural motif present in bioactive compounds and functional organic materials. Despite their importance, the synthesis of complex polyaromatic structures is implemented by customized, target-specific synthetic routes. Unlike peptide and oligosaccharide chemistry, where iterative and automated assembly strategies have transformed discovery, there is no general platform that employs commercially available building blocks for the iterative construction of carbon-carbon bonds. Current state-of-the-art approaches require specialized building blocks, such as bifunctional masked halo-boron reagents.
This Proof of Concept project introduces Pi-LOOP, an iterative synthesis platform for polyaromatic compounds that directly uses commercially available aryl chlorides. The approach exploits π-metal-arene complexation as a multifunctional control element to enable stepwise carbon-carbon bond formation, regioselective C-H borylation, and controlled photorelease of the growing polyaromatic framework. Immobilization of the metal complex on a solid support should facilitate recovery and reuse of the chromium control unit. The project aims to validate the robustness and effectiveness of the Pi-LOOP platform for iterative polyaromatic assembly using off-the-shelf building blocks. By circumventing the need for highly customized reagents, Pi-LOOP can expand polyaryl’s chemical space and open the possibilities toward automation and parallel synthesis. This is relevant to industrial R&D users in pharmaceuticals, agrochemicals, and functional organic materials, where rapid access to structurally diverse polyaromatic scaffolds remains a major bottleneck.
